Blind Super-Resolution Approach for Exploiting Illumination Variety in Optical-Lattice Illumination Microscopy

چکیده
Optical-lattice illumination patterns help in pushing high spatial frequency components of the sample into optical transfer function a collection microscope. However, exploiting these high-frequency require precise knowledge if reconstruction approaches similar to structured microscopy are employed. Here, we present an alternate blind approach that can provide super-resolution without requirement extra frames. For this, property temporal fluctuations emissions using “multiple signal classification algorithm” is extended aptly toward fluctuation phase-modulated lattice illuminations for super-resolution. The ability shown sinusoidal and multiperiodic with approximately 3- 6-fold resolution enhancements, respectively, over diffraction limit.

We here report for the first time the synergistic implementation of structured illumination microscopy (SIM) and multifocus microscopy (MFM). This imaging modality is designed to alleviate the problem of insufficient volumetric acquisition speed in super-resolution biological imaging. SIM is a wide-field super-resolution technique that allows imaging with visible light beyond the classical diff...

Three-dimensional structured illumination microscopy (3D-SIM) is a versatile and accessible method for super-resolution fluorescence imaging, but generating high-quality data is challenging, particularly for non-specialist users. We present SIMcheck, a suite of ImageJ plugins enabling users to identify and avoid common problems with 3D-SIM data, and assess resolution and data quality through ob...

We developed two-photon scanning patterned illumination microscopy (2P-SPIM) for super-resolution two-photon imaging. Our approach used a traditional two-photon microscopy setup with temporally modulated excitation to create patterned illumination fields. Combing nine different illuminations and structured illumination reconstruction, super-resolution imaging was achieved in two-photon microsco...

Structured illumination fluorescence microscopy utilizes interfering light and the moiré effect to enhance spatial resolution to about a half of that of conventional light microscopy, i.e. approximately 90 nm. In addition to the enhancement in the x and y directions, it also allows enhancement of resolution in the z- direction by the same factor of two (to approximately 220 nm), making it a pow...
